Alternative splicing of the platelet-derived growth factor A-chain RNA gives rise to two distinct A-chain forms that differ in the presence or absence of a hydrophilic carboxy-terminus by 18 amino acids (residues 194-211). This peptide, which corresponds to this region and is N-terminally tyrosinated for convenient radioiodination, was shown to bind specifically to several normal and transformed cell lines. Furthermore, this peptide inhibits the stimulation of DNA synthesis and cell division by normal human serum as well as attenuate the growth-promoting activity of several serum growth factors.
