Boc-Asn(Xan)-OH is the asparagine derivitive often utilized in Boc-SPPS. The xanthyl protecting group on the side chain improves the solubility of the asparagine derivative and protects the amide group from dehydration during activation with carbodiimide reagents. The xanthyl protecting group is removed with trifluoroacetic acid (TFA) during Boc deprotection. Since the side chain protecting group is needed mostly during activation and coupling of the asparagine residue, its removal during subsquent Boc removal is not a significant problem. The dehydration side reaction can be avoided by preactivating the amino acid derivatives before coupling or by utilizing activating reagents other than carbodiimides.
