Boc-amino acids were used by Bruce Merrifield in developing solid phase peptide synthesis and they remain popular today. Boc-amino acids are easily dissolved in all of the solvents commonly utilized in solid phase peptide synthesis, including dichloromethane (DCM), N,N-dimethylformamide (DMF), and N-methyl-2-pyrrolidone (NMP). The Boc group is removed with trifluoroacetic acid (TFA), leaving the N-terminus of the peptide protonated.
