Isoacyl Dipeptides
Isoacyl dipeptides are used to prepare long or difficult peptides where aggregation of the peptide chains is prevalent. The isoacyldipeptides are used to synthesize a depsipeptide in which the peptide chain is attached to the side chain O of serine or threonine residues instead of the alpha N. This disrupts inter chain hydrogen bonding responsible for aggregation and improves the solubility of depsipeptides compared to the standard analogs. The depsipeptides are stable under acidic conditions and can be purified by HPLC. When the pH is adjusted to 7.4 or greater, depsipeptides undergo O->N acyl shift to form the native peptide. This provides an efficient way to prepare and purify difficult peptides in high yield.
Guidelines for Utilizing Isoacyl Dipeptides
1. If possible, insert isoacyl dipeptides before hydrophobic regions.
2. There should be at least 2 amino acids between isoacyl dipeptides or between isoacyldipeptides and prolines.
3. The optimum spacing is 5-6 amino acids between isoacyl dipeptides or between isoacyldipeptides and prolines.
