Fmoc-D-Lys(Boc)-OH is the standard lysine derivative for incorporating D-lysine residues into peptides.  The sidechain Boc group of Fmoc-D-Lys(Boc)-OH is removed under acidic conditions such as when the peptide is cleaved from Wang resin or Rink amide resin.

Fmoc-D-Lys(Boc)-OH may be especially useful in preparing new antimicrobial peptides with improved pharmaceutical characteristics.  D-Lysine substituted into selected positions of some antimicrobial peptides can enhance antimicrobial activity, as in macropin (L. Monincová, et al., J. Pept. Sci., 2014, Mar. 11, published online ahead of print) or reduce haemolytic activity, as in  hymenochirin-1B (S. Attoub, et al., Regul. Pept., 2013, 167, 51-6; M. Mechkarska, et al., Peptides, 2013, 50, 153-9).  [D-Lys11] Alyteserin-2a significantly enhances insulin release and glucose tolerance in mice with obesity and insulin resistance (O.O. Ojo, et al., Chem. Biol. Drug Des., 2013, 82, 196-204).  [D-Lys7,11] Alyteserin-2a shows higher potency against Gram-negative bacteria and appreciably lower haemolytic activity than alyteserin-2a (J.M. Conion, et al., J. Pept. Sci., 2012, 18, 207-5).
